PRATZ, J., P. RIPQCHE, AND B. CORMAN. Cholesterol content and water and solute permeabilities of kidney membranes from aging rats. Am. J. Physiol. 253 (Regulatory Integrative Comp. Physiol. 22): R8-R14, 1987.-The influence of the age-related increase in cholesterol content of kidney brush-border membranes on their water and solute permeabilities was studied in loand 24-mo-old male Wistar rats. These permeabilities were determined from the rate of shrinkage and swelling of brushborder vesicles using a stopped-flow apparatus coupled to a light transmission recording. Despite a 20% rise in total cholesterol per milligram proteins, the osmotic water permeabilities of the vesicles were unchanged between 10 (287 t 40 pm/ s) and 24 mo (270 t 31 pm/s). In contrast, passive diffusions of NaCl and KCl, but not of urea, thiourea, and glycerol, were reduced by 35% between 10 and 24 mo. These results suggest that an increase in cholesterol content of kidney brush-border membrane decreases its salt permeabilities without altering the water pathways. cholesterol could be associated with significant permeability modifications. First evidence for changes in solute diffusion consecutive to an age-related rise in cholesterol was reported by Hegner (l2), who found that at between 3 and 22 mo the passive diffusion of cholic acid in rat hepatocytes decreased as plasma membrane cholesterol increased. Unfortunately, comparable data from transporting epithelia responsible for large reabsorptions of solutes and water such as nephron or intestine walls are still lacking.

Application of a fiber-matrix model to transport in renal tubules
The effects of tight junction structure on water and solute fluxes across proximal tubular epithelium were examined with fiber-matrix equations previously derived by Curry and Michel (1980. Microvascular Research. 20:96-99). Using plausible estimates of tight junction fiber length and width the model predicts solute (Ps) and water permeability (Lp) coefficients that agree with the measured valu...
متن کاملWater and solute permeabilities of medullary thick ascending limb apical and basolateral membranes.
The medullary thick ascending limb (MTAL) reabsorbs solute without water and concentrates [Formula: see text] in the interstitium without a favorable pH gradient, activities which require low water and NH3 permeabilities. The contributions of different apical and basolateral membrane structures to these low permeabilities are unclear. We isolated highly purified apical and basolateral MTAL plas...

Integral asymmetric cellulose acetate (CA) membranes were casted by phase-inversion with formamide varying content - 22, 30 and 34% - as pore promoter. These membranes, CA-22, CA-30 and CA-34, were analyzed by infrared spectroscopy in attenuated total reflection mode (ATR-FTIR) to investigate the porous membrane matrix influence on the polymer/water/solute interactions and the selective ultrafi...
متن کاملWater and solute permeability of rat lung caveolae: high permeabilities explained by acyl chain unsaturation.
Caveolae are invaginated membrane structures with high levels of cholesterol, sphingomyelin, and caveolin protein that are predicted to exist as liquid-ordered domains with low water permeability. We isolated a caveolae-enriched membrane fraction without detergents from rat lung and characterized its permeability properties to nonelectrolytes and protons. Membrane permeability to water was 2.85...
